Design Notes

Due to the repeated CDRE, it can be difficult to perform PCR amplification, overlap PCR as well as in-fusion clone. So when standardizing this promoter, we considerately preserved the unrelated sequence. The exact start of this promoter is the first CDRE segment

All the sequences are from Saccharomyces cerevisiae. The segment of CDRE can be found in the article Calcineurin acts through theCRZ1/TCN1-encoded transcription factor to regulate gene expression in yeast[J] and the sequence of CYC1 promoter can be downloaded from NCBI. This fused promoter is synthesized through Gibson Assembly by UCAS.
